Excessive nitrogen or nitrogen deficiency can affect the activity of key enzymes in the metabolism of crops, which will ultimately affect the crop yield. Rice is an important food crop, and nitrogen affected its photosynthesis and photosynthate transport, thereby affecting the yield and quality of rice. This study focuses on the effect of nitrogen fertilizer on RuBP carboxylation enzyme and SPS enzyme activity of rice,in order to explore nitrogen fertilizer on rice growth and the optimal level. The results show that, the RuBP activity in leaves of rice under different nitrogen application was the highest in the blossom period, while decline in the mature period。 low nitrogen were positively related to RuBP carboxylase activity. SPS enzyme activity in a very high content of blossom period, maturity to decline. At the same time, low levels of nitrogen fertilizer led to the lowest SPS activity. These results provide theoretical foundation for the reasonable application of nitrogen in rice growing period.Keywords: rice, nitrogen levels, RuBP carboxylase ,SPS, activity 28
