RTPCR and cloning technology were employed to clone the full coding sequence of IGFBP6 genes out the total RNAs extracted from liver tissue at 0d after birth, followed by using of QPCR to identify the profile of gene expression in different tissues(liver, lung, heart, longissimusdorsi, semimembranosus, musculustriceps) and at different time points(0d、30d、60d、90d、120d after birth). The results indicate: (1) We have cloned the CDS full sequence of IGFBP6 (711bp), which encoding a peptide consists of 235 amino acid residues. Key Words: Nanjiang yellow goat, IGFBP6, Clone, QPCR, Expression1 文獻綜述 IGFBPs的研究進展 IGFs簡介 胰島素樣生長因子(insulinlike growth factors,IGFs)系統(tǒng)在正常生理活動中起著極為重要的作用,其與胚胎分化、個體發(fā)育密切相關,而且參與糖、脂肪和蛋白質代謝。IGFI和IGFII結構與胰島素相似,是兩個同源性較高、分子量大約為7KDa的多肽類激素[1]。IGFs在許多組織中均有表達, IGFs除了通過旁分泌方式發(fā)揮作用外[7],還可以在局部以自分泌的方式促進細胞增殖[8]。IGFs主要通過與IGFI受體結合發(fā)揮促進有絲分裂的作用,IGFI受體與胰島素受體結構相近,均具有受體酪氨酸激酶活性。IGFII受體對IGF的調節(jié)作用目前尚不清楚[9]。IGFs與IGFBPs的親和力高于IGFI受體,因此IGF生物活性和對IGF的代謝與IGFBPs密切相關[10]。IGFBPs是IGFs的重要組成之一,IGFBPs對于調節(jié)IGFs的生物學功能主要表現(xiàn)在以下四個方面: (1)IGFs運載蛋白的作用,并可調節(jié)血液中游離IGFs含量;(2)延長IGFs的半衰期,調節(jié)IGFs的代射清除率;(3)對特異性組織、細胞進行識別和定位;(4)調節(jié)IGFs的生物學效應,通過調節(jié)IGFs和IGFs受體的相互作用來完成;(5)不用依賴于IGFs而直接調節(jié)正常和惡性細胞的生長[1215]。IGFBPs在氨基酸結構上高度保守,在總蛋白質中有35%左右
