中國(guó)礦業(yè)大學(xué) 2020 屆本科生畢業(yè)設(shè)計(jì) 第 1 頁(yè) 年產(chǎn) 300 噸纖維素酶工廠的初步設(shè)計(jì) 中國(guó)礦業(yè)大學(xué) 2020 屆本科生畢業(yè)設(shè)計(jì) 第 2 頁(yè) 摘 要 纖維素是年產(chǎn)量巨大的可再生性資源,地球上每年光合作用生成的上億噸生物質(zhì)中,纖維素占了近一半。目前,自然界中纖維素只有一小部分得到了利用,絕大多數(shù)纖維素不僅被白白浪費(fèi),而且還會(huì)造成環(huán)境污染。利用這一年產(chǎn)量巨大的可再生性資源將其轉(zhuǎn)化為人類(lèi)急需的能源、食物和化工原料,對(duì)于人類(lèi)社會(huì)的可持續(xù)性發(fā)展具有非常重要的意義。 本設(shè)計(jì)采用目前認(rèn)為是最好的產(chǎn)纖維素酶的菌種里氏木霉作為發(fā)酵菌種,液體深層發(fā)酵過(guò)程中采 用變溫發(fā)酵的方法分別控制菌種的生長(zhǎng)和產(chǎn)酶,提取過(guò)程中采用超濾、層析等,提高產(chǎn)品的收率。最后采用噴霧干燥做成固態(tài)的酶制劑。 本設(shè)計(jì)的主要內(nèi)容有:工廠總平面布置、全廠工藝流程設(shè)計(jì)、工藝計(jì)算、設(shè)備的計(jì)算與選型、成本核算;另外,完成設(shè)計(jì)圖紙 8 張,有工廠總平面布置圖、工藝流程圖( 3 張)、發(fā)酵罐設(shè)計(jì)圖、種子罐設(shè)計(jì)圖、發(fā)酵車(chē)間設(shè)備布置圖(平面圖和立面圖)。根據(jù)全廠工藝設(shè)計(jì)和計(jì)算結(jié)果可以看出,該設(shè)計(jì)能夠達(dá)到工業(yè)生產(chǎn)的要求。 關(guān)鍵詞: 纖維素酶; 液體深層發(fā)酵; 里氏木霉 中國(guó)礦業(yè)大學(xué) 2020 屆本科生畢業(yè)設(shè)計(jì) 第 3 頁(yè) ABSTRACT Cellulose is a kind of reproducible resource of great output, it takes about a half of the hundred million biomaterial making by photosynthesis. Presently, only a few cellulose are utilized, most of cellulose are wasted and pollute environment. It is of great importance to transfer these resource to energy ,food, and so on. This design adopt Trichoderma reesei which produce cellulase best. During the liquid submerged fermentation course we chang the temperature in order to control the growth that germ grows and produce cellulase respectively. Ultrafiltration and chromatography are used In the extrace process for improve the yield. In the end we make solid zymin by spray dring . The design mainly include the contents hereinafter: the layout of the whole factory ,the craft argumentation of the whole factory, the calculation of the craft, the calculation and type choosing of main equipments, the calculation of the costs. And design 8 charts , that are the layout of the whole factory, the design of the craft process(3), the design of the fermentation pot, the design of seeding tank, the lay out for equipments of the fermentation workplace(ichnography and space).According to the craft argumentation of the whole factory and the result of the calculation, the design can e up to the request of industrialization.
