【正文】 SHANDONGUNIVERSITY?。希啤。裕牛茫龋危希蹋希牵? 畢 業(yè) 論 文 玉米芯誘導(dǎo)里氏木霉合成纖維素酶學(xué) 院 : 專 業(yè) 班 級 : 學(xué) 生 姓 名 : 學(xué) 號: 指 導(dǎo) 教 師 : 201 年 月摘要I摘要 纖維素類物質(zhì)是自然界中最具潛力的一種可再生資源,其有效利用是目前人們關(guān)注的熱點(diǎn)。作為降解纖維素的多組分酶系,纖維素酶降解纖維素特異性高,反應(yīng)條件溫和,環(huán)境污染小。但由于其組分復(fù)雜,合成代謝網(wǎng)絡(luò)研究尚不清楚。此外,真菌纖維素酶多數(shù)是誘導(dǎo)酶,其合成須在誘導(dǎo)物存在下才能進(jìn)行,通過添加誘導(dǎo)物及對纖維素酶合成途徑進(jìn)行調(diào)控是提高纖維素酶活性的有效方法。因此,研究以天然纖維素物質(zhì)為底物的纖維素酶合成過程機(jī)理,充分挖掘現(xiàn)有生產(chǎn)菌株產(chǎn)纖維素酶的潛力,是解決纖維素酶誘導(dǎo)合成問題的關(guān)鍵。本文對天然纖維素物質(zhì)玉米芯誘導(dǎo)里氏木霉產(chǎn)纖維素酶的過程進(jìn)行了分析,通過測定玉米芯酶解液中糖的組分,并以所測定到的組分為誘導(dǎo)物進(jìn)行誘導(dǎo)產(chǎn)酶,發(fā)現(xiàn)除纖維素成分外,半纖維素水解產(chǎn)物半乳糖對纖維素酶的合成有誘導(dǎo)作用。通過優(yōu)化半乳糖的誘導(dǎo)濃度,發(fā)現(xiàn) 5g/L 的半乳糖對纖維素酶的誘導(dǎo)最好。關(guān)鍵詞:里氏木霉、纖維素酶、玉米芯、誘導(dǎo)AbstractIIAbstractCellulose is a natural substance in the most potential as a renewable resource, and its efficient use is currently the focus of attention.
