Wisconsin Alumni Research Foundation developed a patentable product called Warfarin (from the initials of the foundation + arin to indicate a coumarin pound). VitK拮抗藥 Warfarin – History 1948 51. Warfarin bees a mon 滅鼠劑 (rodenticide) (it still is). 1951. Army inductee tried to mit suicide with Warfarin. He was saved, but the physicians remarked at how good an anticoagulant it was. 1952. Warfarin introduced into clinical use as an oral anticoagulant. VitK拮抗藥 【 體內(nèi)過(guò)程 】 雙香豆素口服吸收慢而不規(guī)則；華法林口服吸收完全。 需監(jiān)測(cè) APTT，保持在 55～ 85 s； 與阿司匹林合用，使 APTT延長(zhǎng) 。 t1/2短。 凝血酶直接抑制藥 阿加曲班 (argatroban) 精氨酸衍生物。 基因重組水蛭素 (rhirudin)作用與天然水蛭素相同 . 預(yù)防血栓 溶栓輔助治療 口服不吸收， ； 不易過(guò)血腦屏障； 原形經(jīng)腎臟排泄，腎衰慎用。 強(qiáng)效、特異的凝血酶抑制劑。 體內(nèi)不代謝，原形尿中排泄。 方達(dá)帕魯（磺達(dá)肝素） 依諾肝素 第一個(gè)上市的 LMWH； 【 藥理作用 】 對(duì)抗因子 Ⅹ a 與因子 Ⅱ 活性比值為 4以上，強(qiáng)大而持久的抗血栓形成； 【 臨床應(yīng)用 】 深部靜脈血栓 預(yù)防外科手術(shù)后靜脈血栓形成 血液透析時(shí)防止體外循環(huán)凝血 【 不良反應(yīng) 】 較少出血，可用魚(yú)精蛋白對(duì)抗 偶見(jiàn)血小板減少 過(guò)敏、肝腎功能障礙禁用 硫酸皮膚素 (dermatan sulfate) 糖胺聚糖類，多聚陰離子； 依賴于肝素輔助因子 (HCⅡ )的凝血酶間接抑制劑 通過(guò)刺激 HCⅡ 通路滅活凝血酶； 提高速率 1000倍。 5. 骨 Ca2+丟失輕。 ? 腎功能衰竭禁用。 2. 吸收更均勻，生物利用度高， tl／ 2更長(zhǎng)， iv可維持
