與下發(fā)揮殺傷病毒感染細(xì)胞的作用。特異性抗體在預(yù)防病毒感染中發(fā)揮重要作用。腺病毒的中和抗原決定簇主要在于六鄰體上,陰性空白組Hela細(xì)胞生長(zhǎng)良好,細(xì)胞呈梭形,排列規(guī)則。而陰性血清組、病毒對(duì)照組Hela細(xì)胞形態(tài)明顯變圓,排列成葡萄串狀,呈典型的腺病毒感染癥狀,試驗(yàn)組在抗體效價(jià)1:50至1:800時(shí)Hela細(xì)胞生長(zhǎng)良好,在抗體效價(jià)為1:1600組開始出現(xiàn)病變,在抗體效價(jià)大于1:3200時(shí),細(xì)胞全部病變明顯。中和抗體試驗(yàn)中血清具有保護(hù)hela細(xì)胞免受腺病毒感染,可以得知其血清中有對(duì)抗腺病毒的成份存在,即重組蛋白具有中和抗原的作用,即antiHx抗體有著極強(qiáng)的中和反應(yīng),證明免疫家兔血清中存在與antiHx抗體作用類似的抗體,進(jìn)一步證明本實(shí)驗(yàn)組所得的重組人腺病毒六鄰體蛋白免疫家兔后產(chǎn)生了預(yù)期的效果。免疫印記實(shí)驗(yàn)表明抗重組六鄰體蛋白血清效價(jià)達(dá)到百萬(wàn)級(jí),說(shuō)明重組人腺病毒六鄰體蛋白具有很高的免疫原性,也證明了所克隆的區(qū)域具有很高的抗原性,與預(yù)期的結(jié)果相一致。而且其高效價(jià)的產(chǎn)生可以在今后的工作中大大提高疫苗的生產(chǎn)效率,為制備特異性強(qiáng)的高效疫苗提供了一定的科學(xué)基礎(chǔ)。從本文的實(shí)驗(yàn)結(jié)果看,免疫家兔后可以誘導(dǎo)實(shí)驗(yàn)動(dòng)物產(chǎn)生高滴度的特異性抗體,并且產(chǎn)生的抗體具有體外中和活性。但是理想的疫苗不僅需要選擇有保護(hù)性的抗原成分, 而且免疫方法、免疫途徑、免疫劑量和免疫方案等對(duì)免疫效果都有直接影響。據(jù)報(bào)道漢坦病毒DNA疫苗用基因槍法注射所產(chǎn)生的抗體滴度比肌肉免疫法高[50]。這要求我們對(duì)載體的選擇, 注射途徑, 注射時(shí)間, 注射劑量等作進(jìn)一步優(yōu)化。本實(shí)驗(yàn)僅證明了重組六鄰體蛋白具有良好的抗原性和3型腺病毒特異性,但該抗體與其他型別的腺病毒是否有交叉反應(yīng)等,都是本課題正在繼續(xù)探討的重要內(nèi)容。結(jié) 論一、證明重組六鄰體蛋白具有腺病毒六鄰體的抗原性。二、證明重組六鄰體蛋白具有腺病毒六鄰體的特異性。其血清抗體效價(jià)高達(dá)1:1 024 000三、重組六鄰體蛋白可以刺激實(shí)驗(yàn)動(dòng)物產(chǎn)生具有保護(hù)作用的中和抗體。參考文獻(xiàn)1.Athappilly FM,Rux JJ.
