批補料發(fā)酵,每升培養(yǎng)液最高可得到50 g干菌種,A600 nm可達125左右。在我們實驗中,嚴格控制補料流加的速度,在有效提高菌體產(chǎn)量的同時提高目的蛋白表達量。高密度發(fā)酵技術(shù)能夠減少培養(yǎng)體積,縮短生長周期,減少設(shè)備投資并強化下游分離提取,為工業(yè)化生產(chǎn)降低成本。本文對重組DH5a/pKKHBLyS的高密度、高表達發(fā)酵研究將可能對其它類型重組蛋白的發(fā)酵生產(chǎn)具有一定的借鑒作用。References: [1] Moore P A, Belvedere O, Orr A, et al.
