【正文】 FE11STV191NAU3942b3討論親本選擇是關(guān)于遺傳連鎖圖譜構(gòu)建的難易程度及圖譜的適用范圍的主要因素之一?;蚪MDNA序列的差異是分子遺傳連鎖圖譜構(gòu)建的基礎(chǔ),因此選擇親緣關(guān)系較遠(yuǎn)的品種或材料作為分離群體構(gòu)建的親本,可以獲得較高的多態(tài)性。與上述方法相比,陸地棉種內(nèi)雜交育種也有重要地位,近年來(lái)在遺傳連鎖圖譜的構(gòu)建的過(guò)程中,有學(xué)者利用陸地棉種內(nèi)雜交群體進(jìn)行。Reddy等[28]比較152對(duì)JESPR SSR引物在異源四倍體棉種的多態(tài)性,%的引物在陸地棉與海島棉種間雜交群體親本TM1與Pima379之間表現(xiàn)出多態(tài)性,%的引物在陸地棉種內(nèi)作圖群體親本TM1和Tamcot SP37中表現(xiàn)出多態(tài)性,%的引物在陸地棉種內(nèi)作圖群體親本HS46和MARCABUCAGBUS188間表現(xiàn)出多態(tài)性。倪會(huì)娟等[29]利用陸地棉品種中棉所35和渝棉1號(hào),從8165對(duì)SSR引物中篩選出335對(duì)多態(tài)性引物,%。王娟等[30]利用陸地棉遺傳標(biāo)準(zhǔn)系TM1和渝棉1號(hào),從5544對(duì)SSR引物中篩選得到178對(duì)多態(tài)性引物,%。但是陸地棉種內(nèi)多態(tài)性較低,與海島棉和陸地棉雜交群體所構(gòu)建的遺傳連鎖圖譜相比,利用陸地棉所構(gòu)建的遺傳連鎖圖譜缺點(diǎn)很多。本研究利用中棉所8號(hào)和Pima9053為親本構(gòu)建的BC1F2海陸雜交群體,以SSR引物構(gòu)建BC1F2的遺傳連鎖圖譜,從190對(duì)具有多態(tài)性的SSR引物中篩選得到262個(gè)多態(tài)性位點(diǎn),最終定位到遺傳連鎖圖譜中的多態(tài)性位點(diǎn)為208個(gè)。構(gòu)建遺傳連鎖圖譜時(shí),研究者都期望標(biāo)記位點(diǎn)能均勻分布于基因組上。自1994年Reinisch等[16]首次報(bào)道四倍體棉花栽培種的RFLP圖譜以來(lái),目前國(guó)內(nèi)外研究者已構(gòu)建了相對(duì)飽和的棉花種間遺傳連鎖圖譜。最為詳盡的遺傳連鎖圖譜是由Rong等[31]利用STS標(biāo)記構(gòu)建的,該圖譜包含2584個(gè)位點(diǎn),26個(gè)連鎖群或染色體。但即便是這樣一個(gè)較高密度的遺傳連鎖圖譜也存在標(biāo)記分布不均勻現(xiàn)象。本研究所構(gòu)建遺傳連鎖圖譜A亞組標(biāo)記多于D亞組,同樣也具有標(biāo)記分布不均的現(xiàn)象。在A亞組9條染色體上分布有110個(gè)標(biāo)記位點(diǎn)。D亞組7條染色體上分布有86個(gè)標(biāo)記位點(diǎn)。同時(shí)標(biāo)記間還存在較大間隙。引起標(biāo)記分布不均的原因是多方面的:第一,研究者篩選到的標(biāo)記可能多數(shù)是由基因組中某一亞組開(kāi)發(fā)得到的;第二,遺傳連鎖圖譜中標(biāo)記數(shù)量較少也會(huì)造成染色體某些區(qū)段標(biāo)記稀疏;第三親本間在染色體某些區(qū)段缺乏多態(tài)性。綜上所述,構(gòu)建遺傳連鎖圖譜過(guò)程中,選用不同種類標(biāo)記和較多的標(biāo)記數(shù)可避免標(biāo)記在基因組上分布不均勻的現(xiàn)象。參考文獻(xiàn):[1] Faerber C.Future demands on cotton fiber quality in the textile industry[R].
