開(kāi)發(fā)產(chǎn)生深遠(yuǎn)的影響.腸道粘膜免疫疫苗對(duì)于腸道粘膜免疫,多以DNA疫苗為主。Tang L.H.等的研究表明,通過(guò)減毒鼠傷寒沙門(mén)氏菌為佐劑的DNA疫苗口服免疫后,可誘導(dǎo)較好的免疫應(yīng)答[22]。Amorij J.P.應(yīng)用流感疫苗,以岍為佐劑消化道免疫小鼠后,增強(qiáng)細(xì)胞輔助性T細(xì)胞的免疫應(yīng)答[23]。另有研究表明。不同佐劑的消化道粘膜免疫.對(duì)疫苗的免疫作用有較大影響。以益生菌基因組DNA為消化道免疫佐劑,削弱了雞對(duì)禽流感病毒H5N1亞型滅活油乳苗的體液免疫應(yīng)答水平,推測(cè)可能與CpG—N、CpG的高度甲基化或者與抗原的分離免疫有關(guān)[24] 。胡青海等以IL一IL一1IFN一cDNA和CpG DNA為佐劑,減毒沙門(mén)氏菌運(yùn)送H5亞型禽流感DNA疫苗消化道和鼻腔聯(lián)合免疫,結(jié)果表明,CpG佐劑組攻毒保護(hù)指數(shù)最高[25]。不同的免疫途徑相結(jié)合.可誘導(dǎo)較好的粘膜免疫應(yīng)答,以前的研究也證明了這一點(diǎn)。腸道粘膜免疫載體脂質(zhì)體作為疫苗載體,具有免疫佐劑效應(yīng)。Locher等[26] 研究表明,用脂質(zhì)體包裹編碼HIV2 ENV gpl40基因的DNA疫苗,免疫小鼠3次后,誘發(fā)了高水平的系統(tǒng)IgG和粘膜IgA抗體反應(yīng)。細(xì)菌栽體主要包括李斯特桿菌、鼠傷寒桿菌、志賀菌、乳酸桿菌屬等減毒細(xì)菌用重組李斯特桿菌作為HIV Gag疫苗載體,經(jīng)直腸、靜脈、口服等途徑免疫小鼠,在脾和腸相關(guān)淋巴組織誘發(fā)HIV Gag特異性CD8+T細(xì)胞[28] 。疫苗以沙門(mén)氏菌為載體,表達(dá)SIV Gag蛋白和沙門(mén)氏菌Ⅲ型分泌性sopE蛋白融合體,口服初免獼猴,以嵌有SIVmac239 Gag的重組痘苗病毒(MVA Gag)皮內(nèi)注射加強(qiáng)免疫,可誘發(fā)SIV特異性CTL反應(yīng)。[29]聚乙烯亞胺PEI等陽(yáng)離子聚合物載體與其他形式的基因載體相比具有下述優(yōu)點(diǎn):無(wú)免疫原性,不會(huì)引起機(jī)體的免疫反應(yīng);遺傳物質(zhì)的釋放可做到由聚合物基質(zhì)的降解速率決定。PEI已經(jīng)證明是有效的、毒性小、適用于許多免疫途徑的基因轉(zhuǎn)移載體,在基因治療和生物學(xué)研究領(lǐng)域有著廣闊的應(yīng)用前景。[3032]參考文獻(xiàn)[l]Royce RA,Sena A,Cates W,et al Sexual transmission of HIV[Jj N Engl J Med,1997,336:1072 1078.
