【正文】 thod (MTT). the concentration of BALP and OC of osteoblasts were determined by ELISA different time points (12h, 24h, 48h, 72h) after optimal dosage of noggin BMP2, IGFI were added into the culture the expression level of BALP and OC were investigated by reverse transcription PCR at the same time. Results: 1. The proliferation ability of MG63 cells treated with BMP2, noggin and IGFI groups were significantly different pared with that of CON group(P<). The effects of three factors on MG63 cell was dosedependently and timedependently after cultured for 72h. The most effective growth of MG63 cells showed at 2109 mol/L of BMP2 with the proliferation rate of ℅ and 1108mol/L of IGF1 with the rate of ℅, while 8109mol/L noggin with the inhibitory proliferation rate of ℅, respectively. 2. The concentration of BALP treated with BMP2, noggin and IGFI groups were significantly different pared with that of CON group (P< ) for 48h, especially, the noggin group was extremely significant (P< ), and the BALP mRNA expression were also significantly different (P< ). 3. The concentration of OC treated with noggin groups were significantly different pared with that of CON group (P< ) for 24h, but the OC mRNA expression were not significantly different (P> ). Key words: Noggin。 bone morphogeic protein 2 (BMP2)。 insulinlike growth factor I (IGFI)。 MG63 cell。 proliferation。 differentiation. 前 言 生長是動物個體發(fā)育特有的生理特點,其主要表現(xiàn)在體高體長的增長方面。而體高體長的增長則源于骨骼的縱向生長,從而形成相應(yīng)的骨骼系統(tǒng),達到體高體長的增長的目的。骨骼系統(tǒng)也是機體非常重要的一個組成部分,它支撐軀體,保護內(nèi)臟,并且和骨骼肌一起協(xié)調(diào)配合完成其運動功能。 在骨骼的生長發(fā)育過程中,骨組織不斷進行著新骨形成和舊骨吸收的骨重建過程,即骨形成( bone formation)與骨吸收( bone resorption)的動態(tài)平衡代謝過程。骨形成由成骨細胞介導(dǎo)來完成。在骨形成過程中,成骨細胞主要經(jīng) 歷細胞增殖、細胞外基質(zhì)成熟、細胞外基質(zhì)礦化和細胞凋亡四個階段。在幼年生長時期,骨形成遠遠大于骨吸收 ( Krupa B et al, 2020) ,為骨骼的生長提供了良好的基礎(chǔ),也為體高體長的增長提供了重要的保證。在成年時期,骨的形成較為穩(wěn)定,骨骼的生長不再發(fā)生變化,這主要是受到骨形成與骨吸收相對均衡的一個動態(tài)調(diào)節(jié) 作用 來完成。在晚年時期,則主要趨向于骨吸收大于骨形成,從而骨形成會受到一定的影響,這將導(dǎo)致一系列骨疾病的發(fā)生,比如在老年人群體中常出現(xiàn)的骨質(zhì)疏松癥。 然而 骨骼的縱向生長是一種十分復(fù)雜的過程,是多 種內(nèi)分泌激素、生長因子在時空上對生長板綜合作用的結(jié)果。內(nèi)分泌系統(tǒng)的全身性激素如生長激素、降鈣素、甲狀旁腺激素、性激素、糖皮質(zhì)激素等,與生長因子如胰島素樣生長因子 ( Iinsulinlike growthfactorI, IGFI)、骨形態(tài)發(fā)生蛋白 (bone morphogeic proteins, BMPs)、轉(zhuǎn)化生長因子 (transforming growth factor TGF)、成纖維細胞生長因子 (fibroblast growthfactors, FGF)、血管內(nèi)皮生長因子( vascular endothelial growth factor, VEGF)、甲狀旁腺素相關(guān)肽( PTHrelated peptide, PTHrP)等局部因子,以及外源性物質(zhì)如維生素 D 等相結(jié)合,共同調(diào)節(jié)骨骼生長過程 。 1.
