【正文】 cr擴(kuò)增擴(kuò)增反應(yīng)體系:10pcr緩沖液5 μl,mgcl2(25 mmol/l) 4 μl,dntp 2 μl,上下游引物各1.5 μl,模板dna 1 μl,taq酶 (10 000 u/ml) 0.5 μl,加去離子水至50 μl。1 材料與方法1.1 供試土壤供試土壤取自鹽城海濱濕地土壤,無人為污染。結(jié)果表明,各處理下的土壤細(xì)菌群落多樣性在不同處理時間有明顯差異。關(guān)鍵詞:變性梯度凝膠電泳(dgge);對二氯苯;濕地土壤細(xì)菌菌群;多樣性abstract: the effect of different concentration of 1,4-dichlorobenzene(120,240,360 and 600 mg/kg) with different cultivation time (4, 18 and 45 d) on wetland bacterial community diversity was studied using pcr-dgge technique. the results showed that the effect of each concentration of 1,4-dichlorobenzene on genetic diversity of soil bacteria differed at different treating time. in early treatment period, soil bacterial community was inhibited obviously. the difference among different treatments in later period was little. in early treatment period, a new medium band emerged in the culture medium of 240 mg/kg of 1,4-dichlorobenzene treatment, indicating that there were special microbial species resistant to 1,4-dichlorobenzene existed in the soil.key words: dgge; 1,4-dichlorobenzene; wetland soil bacteria communities; diversity土壤微生物在土壤生態(tài)中扮演了重要角色[1],并且參與地球生物生態(tài)系統(tǒng)的化學(xué)循環(huán),進(jìn)行污染物質(zhì)的降解[2,3]。桶內(nèi)加入配制好的對二氯苯母液,使桶內(nèi)對二氯苯濃度分別為0、120、240、360和600 mg/kg,每組3次重復(fù),室溫20 ℃培養(yǎng)。反應(yīng)結(jié)束后取5 μl反應(yīng)液在1%的瓊脂糖凝膠上進(jìn)行電泳檢測。由圖1可知,試驗初期,不同濃度對二氯苯脅迫下濕地土壤細(xì)菌群落的基因條帶出現(xiàn)較明顯的差別。由圖3可知,在處理18 d時,與對照相比,120 mg/kg對二氯苯脅迫與對照土壤的細(xì)菌群落有77.4%的相似性,再
