Chen)又名花梨木DNA 條形碼測定中的有效性。從基地采摘的降香黃檀葉片中提取 DNA,用特 異性引物進行 PCR 擴增,得到三條候選條形碼的目的片段,擴增成功的 DNA 片段可用于進一步測序分析。以 PCR 擴增成功率和測序成功率初步評價候選 DNA 條形碼片段,評選出最優(yōu)的條形碼片段,為今后利用 DNA條形碼鑒定木材和構(gòu)建木材分子條形碼數(shù)據(jù)庫提供理論依據(jù)。 實驗結(jié)果:只有 ITS2 的條形碼片段測序成功,得到對應(yīng)的序列。通過NCBI 查詢黃檀屬其他物種的 ITS2 序列,得到:降香黃檀的 ITS2 分子序列與黃檀屬植物均不同,其中與越南黃檀差異最小,其次是多裂黃檀。多重比對后構(gòu)建該序列的進化樹結(jié)果說明降香黃檀與越南黃檀、多裂黃 檀有著較近的親緣關(guān)系,但又不屬于同一植物類群,且三者有著穩(wěn)定的遺傳差異性。建議把 ITS2 序列作為鑒定降香黃檀的候選條形碼序列。 關(guān)鍵詞: 降香黃檀; DNA 條形碼; ITS2;分子鑒定 降香黃檀 DNA 條形碼序列測定 II II Abstract DNA barcode as the geic materialin life inherent that objectively existed in the cell, without artificial interference. It could accurately reflect the most essential information for species. In order to verify three candidate barcodes of the MatK, trnH – psbA and ITS2 remended by CBOL and the effectiveness in Dalbergia odorifera ’s DNA barcode. Extracted DNA of its leaves from the base,using specific primers for PCR amplication. The purpose of the three candidates from bar code fragments were obtained and the targeted segment was amplified successfully to be used for sequencing analysis. PCR amplification and sequencing success rate could be used for the preliminary evaluation the bar code fragments of candidate DNA ,Selected the best bar code fragment,which provided theoretical basis for using DNA identification of wood and wood construction molecule. The results were as follows : only ITS2 coded fragment was successfully sequenced. and the corresponding sequence was gained. By searched ITS2 of other species which belong to Dalbergia from NCBI. We got the consion conclusion that: ITS2 of Dalbergia odorifera were different,with Dalbergia tonkinensis minimum, followed by Dalbergia rimosa Multiple parison after building the sequence of the evolutionary tree result showed that Dalbergia odorifera Dalbergia tonkinensis and Dalbergia rimosa had the close relationship. Though they are not belong to the same group of plants, and they have a stable geic diversity. So ITS2 sequence can be uesd to identified candidate barcode of Dalbergia.
