的分解纖維素的微生物。細(xì)菌適宜在酸堿度接近中性的潮濕土壤中生長(zhǎng),絕大多數(shù)分布在距地表約 3— 8cm 的土壤層。本次試驗(yàn)就是的目的就是從土壤中將這些能夠分解纖維素的細(xì)菌分離出來,并對(duì)它們的形態(tài)特征、生理生化等進(jìn)行研究和探討。地球上的植物每年產(chǎn)生的纖維素超過70 億噸,其中 40%— 60%能被這些微生物分解利用。因此,開發(fā)高效轉(zhuǎn)化木質(zhì)纖維素類可再生資源的微生物技術(shù),將生產(chǎn)生活中產(chǎn) 生的工農(nóng)業(yè)廢棄物轉(zhuǎn)化為可利用的新能源,將具有極其重要的意義和廣闊的前景。對(duì)于人類來說,生物質(zhì)資源是寶貴的可再生資源,是我們賴以生存的基本物質(zhì)資源。 關(guān)鍵詞 土壤 纖維素分解菌 分離篩選 pH 試驗(yàn) Isolation and Screening of Soil CelluloseDeposing Microbes Zhou Yudi, Zhu Liang, Zhu Xiaoyu, Zong Lu School of Life Science, Shandong University, Jinan,250100 Corresponding author Abstract In our experiment, we took an appropriate amount of soil sample, dissolved them in distilled water, put them in enrichment culture, and then sifted the bacteria and fungus which can break down cellulose by Congo redcellulose medium. After that, we did the pure cultures to observe the color, shape and some other characteristics of the colonies, moreover, we stained the bacteria we got with Gram stain. Later we did some physiological and biochemical experiments such as methyl red test, amyl hydrolysis experiment and pH gradient test to see whether these bacteria can produce acid, whether they can hydrolyze starch and their ability of breaking down cellulose under different PH valves. We hope that by doing this experiment, we can lay the foundation for further research in the future.
