[周志湘, 萬方浩, 張桂芬, 陳斌, 2007. 煙粉虱基因組DNA快速提取方法. 植物保護(hù), 33(5): 131133](責(zé)任編輯:袁德成)。如Moritz (2000)采用轉(zhuǎn)錄間隔區(qū)(internal transcribed spacer, ITS)限制片段長度多態(tài)性(ITSRFLP)的方法,先行擴(kuò)增出西花薊馬和美棘薊馬 Echinothrips americanus Morgan的ITS1和ITS2片段,然后選用 5 種內(nèi)切酶消化兩種薊馬的ITS片段,以產(chǎn)生不同的帶型;此方法可以成功地將兩種薊馬區(qū)分開,但是無法將西花薊馬與其他種類的薊馬區(qū)分開。西花薊馬體型微小, mm左右,~ mm,而且與其他常見的薊馬種類如花薊馬、禾花薊馬、煙薊馬等形態(tài)相似;并且,成蟲常將卵產(chǎn)在寄主植物組織中,而預(yù)蛹期和蛹期主要在土壤中度過。. 1: 滿天星花瓣Flowers of baby39。 2: 1齡幼蟲First 1st instar larva。 西花薊馬 SCAR引物的種特異性及靈敏性檢驗(yàn)以西花薊馬及田間采集的其他41種薊馬的DNA為模板,以所設(shè)計(jì)的特異性引物FOMF/FOMR進(jìn)行PCR擴(kuò)增,電泳檢測結(jié)果顯示(圖2),該引物只對西花薊馬具有擴(kuò)增效果,對其他種類的薊馬不具有擴(kuò)增能力,表明該引物為西花薊馬的特異性引物。 9: 茶黃硬薊馬Scirtothrips dorsalis Hood。 1: 西花薊馬F. occidentalis(Pergande)。C℃ 45 s;最后 72 176。擴(kuò)增反應(yīng)在 ABI9700 PCR基因擴(kuò)增儀上運(yùn)行。然后分別采用上述6 條隨機(jī)引物(上海生工生物工程技術(shù)有限公司協(xié)助合成),以西花薊馬以及其他 9 種(即花薊馬 F. intonsa、禾花薊馬 F. tenuicornis、煙薊馬 T. tabaci、蔥韭薊馬 T. alliorum、黃胸薊馬 T. hawaiiensis、八節(jié)黃薊馬 T. flavidulus、豆大薊馬 Megalurothrips usitatus、茶黃硬薊馬 Scirtothrips dorsalis 和稻簡管薊馬 Haplothrips aculeatus)田間常見薊馬種類的DNA為模板,進(jìn)行PCR擴(kuò)增。C℃ 12 000 r/min 離心 10 min,取上清液,加入 440 μL預(yù)冷無水乙醇,輕柔混勻后于 30 176。該研究結(jié)果為西花薊馬快速檢測體系的構(gòu)建提供了依據(jù),為有效阻截西花薊馬在我國的進(jìn)一步擴(kuò)散蔓延和危害提供了技術(shù)保障。近年來分子生物學(xué)技術(shù)發(fā)展迅速,利用分子標(biāo)記技術(shù)只需要少量的DNA就可以快速準(zhǔn)確地的鑒定出薊馬種類,該類技術(shù)不僅可以檢測成蟲,對成蟲前的各個蟲態(tài)亦具有同樣的檢測效能,目前已應(yīng)用于西花薊馬的鑒定研究(Moritz et al., 2000。即使在非適生區(qū),隨著溫室、大棚等設(shè)施農(nóng)業(yè)的快速發(fā)展,西花薊馬的發(fā)生也會成為可能。 3.
